Issac Hitchcock Jr. was born on April 4, 1817, in Kentucky. He was the youngest child of Issac Hitchcock and Elizabeth Wheeler. Issac had at least seven siblings: Joshua Hitchcock (born 1799), Jesse Hitchcock (born 1801), Sampson Hitchcock (born 1908), Silas Marion Hitchcock Sr. (born 1807), Absalom Hitchcock (born 1810), Elizabeth Isabella Hitchcock (born 1812), and Lydia Hitchcock (born 1814).
Issac married Elisabeth Ross on February 15, 1835, in Jackson County, Missouri. Issac and Elizabeth were married for fifty-five years and had at least eight children between 1835 and 1858: John Green Hitchcock (born 1838), Elizabeth Frances Hitchcock (born 1838), Andrew P Hitchcock (born 1840) William Franklin Hitchcock (born 1847), Parthenia Jane Hitchcock (born 1849), Charles Webber Hitchcock, born 1854), Issac Allen Hitchcock (born 1856), and James Madison Hitchcock (born 1858).
